You would've filled in the bg, and left the squares blank. Open up Jasc Animation shop.

Because there are 5 squares, I want 5 frames. So what I do is I go to animation>insert frames>from file. Select Add File, and I select the image that I was using before. I do this 4 times to get 5 frames. I then resize my box so i can see all my frames.

Step 2.

I pick the Paint Bucket tool and in each frame, select a different box and fill it in with the colour of your choice. Ill do dark green so you can see it.

Step 3.

That's it!!! Now you just set the frame speed to what you want (Alt+Enter or Animation>Frame Properties) and set the animation properties (Shift+Alt+Enter or Animation>Animation properties), looping to "Repeat the animation Indefinately". This is what I got.
